dc.description.abstract | S. cerevisiae'da glukoz tercih edilen bir karbon ve enerji kaynağıdır. S. cerevisiae'da glukozun hücre içine taşınması hücre membranında bulunan Hxt proteinleri tarafından yapılır. HXT4 geni transkripsiyonu glukoz sinyali ile kontrol edilir ve glukoz taşınmasında önemli işlevi vardır. Hücre içine taşınan glukoz ise hücrenin fizyolojik durumuna göre metabolize edilir. Abiyotik stres koşullarında S. cerevisiae'da stres koruyucu metabolitler olarak glikolitik yola bağlı olarak trehaloz ve gliserol sentezlenmektedir. Bu araştırmada HXT4 transkripsiyonuna abiyotik stres koşullarının etkileri incelendi. Ozmotik stresin HXT4 geni transkripsiyonunu baskıladığı görüldü. Oksidatif stresin ise HXT4 geni transkripsiyonunu aktive ettiği belirlendi. Diğer taraftan, sıcaklık stresinin ise belirli koşullarda HXT4 geni transkripsiyonunu aktive ettiği bulundu. Bu sonuçlara ek olarak stres koşullarında aktive edilen transkripsiyon faktörleri Hot1p ve Yap1p'nin HXT4 geni transkripsiyonuna etkilerinin olmadığı bulundu. | tr_TR |
dc.description.abstract | Glucose is the preferred carbon and energy resource in S. cerevisiae. Glucose transport into the cell is carried out by the Hxt proteins localized in the cell membrane. Transcription of the HXT4 gene is regulated by glucose signaling and it has a significant function in glucose transport. Once transported into the cell, glucose is metabolized depending on the physiological status of the yeast cell. Under abiotic stress conditions, trehalose and glycerol are synthesized from glycolytic intermediates as stress protectant metabolites in S. cerevisiae. In this research, effects of abiotic stress conditions on the transcription of HXT4 were investigated. It was found that while osmotic stress represses the transcription of HXT4, oxidative stress activates its transcription. On the other hand, it was shown that the temperature stress also activates the transcription of HXT4 gene in certain conditions. In addition to these results, it was found that the stress activated transcription factors Hot1p and Yap1p have no effect on the transcription of HXT4 gene. | en_US |
